Dialysable transfer factor (TF-D) was prepared from human buffy coat cells and chromatographed on Sephadex G-25 and DEAE cellulose. Column fraction pools enhanced in vitro lymphocyte proliferation induced by the antigen streptokinase-streptodornase. In contrast to the discrete localization of bed-adherent small molecule standards eluted from Sephadex G-25, bed-adherent enhancing activity was dispersed in many fractions. These results and others, showing elution of activity from DEAE cellulose at more than one salt concentration, suggest that activity results from the action of chemically heterogeneous low molecular weight (dialyzable) molecules.
